What is Friedel Crafts Reaction?

Friedel-Crafts Reaction is a fundamental class of reactions in organic chemistry used to introduce alkyl or acyl groups into an aromatic compound. Discovered by Charles Friedel and James Crafts in 1877, these reactions are widely used in the synthesis of aromatic hydrocarbons, ketones, and other derivatives for applications in the pharmaceutical, agrochemical, and polymer industries.

Friedel Crafts in the Synthesis of Ibuprofen

The Friedel-Crafts acylation reaction is a key step in the industrial synthesis of Ibuprofen, a widely used non-steroidal anti-inflammatory drug (NSAID). This reaction is employed in the Boots synthesis route, one of the most efficient commercial processes for producing Ibuprofen.

Reaction

Aromatic Substrate: Isobutylbenzene

Acylating Agent: Acetyl chloride (CH₃COCl)

Catalyst: Aluminum chloride (AlCl₃) (acts as a Lewis acid)
